Lenti-CHIKV E3/E2/6k/E1 Pseudovirus [Luciferase, VSV Packaging System]

The 293T cells expressing CHIKV E3/E2/6k/E1 proteins were infected with VSVΔG* which encodes the luciferase gene. After 24 h the infected cell supernatants were collected and the pseudotyped viruses were purified by centrifugation through sucrose density gradients. The produced pseudoviruses carry CHIKV E3/E2/6k/E1 proteins, which is a powerful tool for vaccine development and entry inhibitor research.

Target Details

Introduction : Chikungunya virus (CHIKV) is an alphavirus belonging to the Togaviridae family. It has a positive-sense single-stranded RNA genome with 11.8 kb long. The genome encodes five structural proteins, the capsid and the envelope proteins (E3, E2, 6k, and E1). E2 and E1 form a heterodimer that projects as an outward-facing spike on the surface of CHIKV particles. E2 protein regulates the virus attachment to cell receptor and E1 may be responsible for promoting the release of the viral nucleocapsid. E3 glycoprotein is associated with mature virions in some alphaviruses. 6K protein is incorporated into particles at a low level andnappears to potentially have multiple roles in glycoprotein processing, cell permeabilization, and viral budding.
