Lenti-SARS-CoV Spike Pseudovirus [GFP, HIV-1 Packaging System]
SARS-CoV spike pseudovirus is produced by cotransfecting HEK293 cells with the plasmid encoding S-protein variants and the plasmid containing the SIV genome with the env gene deleted and the nef gene replaced by that for GFP. Pseudovirus-containing supernatant was harvested 48 h following transfection. The supernatant was cleared of cell debris by centrifugation and filtered through a 0.45-μm-pore-size filter.
