Lenti-EBOV GP Pseudovirus [EGFP, VSV Packaging System]
EBOV GP Pseudovirus is produced by cotransfecting 293T cells with pcDNA3.1 plasmid expressing EBOVΔO GP using the calcium phosphate transfection procedure. Cells were rinsed with phosphate-buffered saline (PBS) 12 h later to remove the transfection reagents. At 24 h following transfection, cells were transduced with VSVG pseudotyped VSV G-eGFP. Viral inoculum was removed 12 h later, and supernatants were collected at 24 h following transduction for viral stocks. The supernatant was cleared of cell debris by centrifugation and filtered through a 0.45-μm-pore-size filter. The produced pseudoviruses carry EBOV GP proteins, which is a powerful tool for EBOV glycoprotein function and entry inhibitor research.

Introduction : | Ebola viruses (EBOVs) are the cause of repeated outbreaks of fatal infections, including the recent deadly epidemic in West Africa. The EBOV has a membrane envelope decorated by trimers of a glycoprotein (GP) which is cleaved by furin to form GP1 and GP2 subunits). GP is solely responsible for host cell attachment, endosomal entry and membrane fusion, thus becoming a primary target for the development of antiviral drugs. |
