Human metapneumovirus (HMPV) belongs to the Pneumoviridae family and Metapneumovirus genus. HMPV is a negative-sense single-stranded RNA virus and is pleomorphic in nature and its size varies from 150 nm to 600 nm. It is a ubiquitous respiratory pathogen that has been circulating in human populations undetected for decades until hMPV was isolated in 2001. Since then, hMPV has been detected in 4-16% of patients with acute respiratory tract infection (ARI). Globally, HMPV is recognized as the second most common cause of bronchiolitis and pneumonia in children <5 years of age. Based on monoclonal-antibody studies and the sequencing of multiple genes, hMPV exists as two genotypes, A and B, that are further subdivided into lineages A1, A2, B1, and B2. Subgroup A2 is again subdivided into A2a and A2b.
The attachment glycoprotein (G), fusion protein (F) and small hydrophobic protein (SH) of the virus protrude out on the virion surface while matrix protein (M) lines the inner surface of the lipid bilayer envelope. The nucleoprotein (N), Phosphoprotein (P) and RNA-dependent RNA polymerase (L) wraps the negative-sense single-stranded RNA genome in the viral core and constitute the ribonucleoprotein (RNAP) complex. HMPV G is capable of binding cell surface GAGs and may contribute to virus attachment. HMPV F is necessary and sufficient for membrane fusion and capable of mediating entry without an additional attachment protein. The proteins matrix 2 proteins, M2-1 and M2-2, are expressed only during the replicative viral cycle. G, M2-1, M2-2 and SH proteins may be involved in the evasion of the immune response.
