Lenti-hRSV SH/G Pseudovirus [GFP, HIV-1 Packaging System]

Lenti-hRSV SH/G Pseudovirus is produced by cotransfecting 293T cells with the hRSV envelope protein expression construct pcDNA-hRSV-SH and pcDNA-hRSV-G, the pCMV-ΔR8.74 lentiviral Gag-Pol expression construct, and a GFP transducing vector (pWPI_Venus-GFP_BLR). Pseudovirus-containing supernatant was harvested 30 h following transfection. The supernatant was cleared of cell debris by centrifugation and filtered through a 0.45-μm-pore-size filter. The produced pseudoviruses carrying hRSV SH/G is a powerful tool for studying mechanism of virus entry, antiviral mechanism, neutralization antibody assay, and screening antivirus inhibitor.

Target Details

Introduction : Human respiratory syncytial virus (hRSV) is an enveloped, nonsegmented, negative-strand RNA virus classified in the family Paramyxoviridae. It causes upper and lower respiratory tract disease in children and elderly and immunosuppressed persons. hRSV possesses three envelope proteins, the fusion glycoprotein (F), the attachment glycoprotein (G), and the small hydrophobic protein (SH). The F protein directs membrane fusion independently of other viral envelope proteins. The hRSV-G protein is highly variable and heavily glycosylated. It is important for virus attachment, primarily through interaction with glycosaminoglycans. The hRSV-SH protein forms oligomeric channel-like structures with 5- or 6-fold symmetry with a nonselective cation channel activity in mammalian cells.
