Retro-HIV-1 DJ263.8 gp160 Pseudovirus [Luciferase, HIV-1 Packaging System] (CAT#: PSV-012CQ)

HIV-1 gp160 pseudovirus is produced by cotransfecting 293T/17 cells with a plasmid inserted an HIV-1 env gene derived from DJ263.8 isolate and an env-deficient HIV-1 backbone plasmid using Lipofectamine 2000 (Invitrogen). Pseudovirus-containing supernatant was harvested 24 h following transfection. The supernatant was cleared of cell debris by centrifugation and filtered through a 0.45-μm-pore-size filter. The produced pseudoviruses carry HIV-1 gp160 proteins, which is a powerful tool for HIV-1 vaccine and entry inhibitor research.

IntroductionHuman immunodeficiency virus (HIV) can be divided into two major types, HIV-1 and HIV-2. HIV-1 is the most common and pathogenic strain of the virus. The HIV-1 env gene encodes the gp160 glycoprotein, which is subsequently cleaved into the envelope proteins gp120 and gp41. gp120 is the viral surface protein that mediates attachment of the virus to target cells via binding to the CD4 receptor and a co-receptor such as CCR5. gp41 is a transmembrane protein that mediates fusion of the viral and cellular lipid membranes once gp120 has bound its cognate receptor and co-receptor.
