Introduction | The MERS-CoV spike (S) protein is a type I transmembrane glycoprotein which contains 1353 amino acids and can be cleaved into two subunit S1 and S2. It forms large protruding spikes on the surface of the virus. The S1 subunit which contains RBD is responsible for binding to Dipeptidyl peptidase 4, which serves as the host cell receptor of MERS-CoV and S2 mediates the membrane fusion. In this process, heptad repeats 1 and 2 (HR1 and HR2) of the S protein assemble into a complex called six-helix bundle (6-HB) fusion core structure, which represents a key membrane fusion architecture. Importantly, the S protein is considered a key component of vaccines against coronavirus infection, including severe acute respiratory syndrome (SARS). |
