SARS-CoV spike pseudovirus is produced by cotransfecting HEK293 cells with the vector pHxLacZWP encoding -galactosidase (-Gal), pShCAG2-nSpike, and HIV packaging plasmid pCMVR8.2. Pseudovirus-containing supernatant was harvested 48 h following transfection. The supernatant was cleared of cell debris by centrifugation and filtered through a 0.45-μm-pore-size filter. The produced pseudoviruses carry SARS-CoV spike protein, which is a powerful tool for SARS-CoV vaccine and entry inhibitor research.
