Human CD4-overexpressed 293T pseudohost is produced by transfection 293T cells with a lentiviral vector encoding human CD4 proteins. This stable cell line express CD4 proteins on the cell surface, and it can be used for drug/vaccine/viral inhibitor screening, neutralizing antibody screening and serum neutralization assay.

|Mycoplasma Testing||The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.|
|Quality Control||Cultures are screened for the presence of bacteries, yeast, fungi and mycoplasma (DNA amplification). Growth media are also certified based on U.S. Public Health Service Guidelines.|
|Storage||Cells must be thawed immediately upon receipt and grown according to handling procedures to ensure cell viability and proper assay performance. DO NOT FREEZE the cells upon receipt as it may result in irreversible damage to the cell line. After recovery, the cells can be stored at -80°C or liquid nitrogen (-150°C to -190°C) for long term.|

|Introduction||CD4 (cluster of differentiation 4) is a glycoprotein found on the surface of immune cells such as T helper cells, monocytes, macrophages, and dendritic cells. It is a member of the immunoglobulin superfamily. HIV-1 uses CD4 to gain entry into host T-cells and achieves this through its viral envelope protein known as gp120. The binding to CD4 creates a shift in the conformation of gp120 allowing HIV-1 to bind to a co-receptor expressed on the host cell. These co-receptors are chemokine receptors CCR5 or CXCR4. Following a structural change in another viral protein (gp41), HIV inserts a fusion peptide into the host cell that allows the outer membrane of the virus to fuse with the cell membrane.|
